A cadre of European politicians gathered Thursday at the Museum of the 18th century in Brussels
to launch Europeana, a digital
museum that allows visitors to explore classic paintings, photos, recordings and texts in the
same manner in which it is possible to search, say, Amazon.com.
Trying to access Europeana on the day of its launch, though, was akin to navigating the Vatican
Museums in the tourist-thick month of August. It was impossible to see anything, as the
project’s three servers were totally overwhelmed.
The Commission said Saturday in a press release that the site received about 10 million hits per
hour throughout Thursday - double server capacity. The site was taken down Friday evening and is
expected to be back up in mid-December.
Europeana’s three servers are located in the Hague, where the project is headquartered, but
programmers plan eventually to put mirror servers around the world.
A pair of Dutchmen programmed Europeana in about 10 weeks, said technical developer Eric Van der
Meulen. They added the final two of 21 European languages, Finnish and Hungarian, at 7 p.m. on
Wednesday.
Europeana, which is still in beta, was programmed using only open source applications, Van der
Meulen said.
“Once we get the thing finished and stabilized, we want to be able to put this down as an
open source application so other people can look at it and go, ‘Ok how did you do
this?,’ and ‘Wow, maybe we can use this for something.’ The future of computing
is open source and not only that but you can get a lot of input from all over the world this
way.”
Technical challenges included harvesting and normalizing metadata from more than 1,000 different
museums and libraries from around Europe. Half of participating cultural heritage institutions so
far are French. The Louvre in Paris, the Institut National de l’Audiovisuel (which
contributed footage shot on French battlefields in 1914) and the Rijksmuseum in Amsterdam are
three of the biggest participating museums.
Europeana is an outgrowth of The
European Library, on which Van der Meulen also worked. But it has in the press been compared
to Google’s Library
Project. Copyright concerns are abundant in all three projects.
Viviane Reding, European commissioner for media, worked to bring the European Digital Library to
fruition prior to realizing Europeana.
Issues of intellectual property will certainly complicate Reding’s goal of adding 10
million more objects over the next two years. The project will receive 2 million Euro over the
next two years for that goal, said European Commission president Jose Manuel Barroso on Thursday.
For now, all objects on Europeana are in the public domain.
Reding said Thursday that she encourages users of the site to ‘remix’ what’s
available. Moving forward, she plans to facilitate dialogue among various stakeholders to find a
way to legally include contemporary works. Nobody wants a black hole when it comes to artifacts
from the 21st century, she stated. In particular, she said she will continue discussions with
books publishers in order to arrange for digitization of orphan works.
The difference between Europeana and existing library projects, though, is in the diversity of
digital objects available on Europeana. Van der Meulen, for example, is able to search the names
of his family members and come to a recording of his uncle’s 1970s rock band, the Makkers,
or photos of his father Leendert Van der Muelen, a world-class cyclist.
“It’s for a lot of people that way,” he said. “Its a fun toy. Everybody
Googles their name, you know. Only with this you get associations with your own name that you
wouldn’t find in Google.”

selectively cleave proteins in response to calcium signals and thereby control cellular functions
such as cytoskeletal remodelling, cell cycle progression, gene expression and apoptotic cell death.
In mammals, the two best-characterized members of the calpain family, calpain 1 and calpain 2
(micro-calpain and m-calpain, respectively), are ubiquitously expressed. The activity of calpains
is tightly controlled by the endogenous inhibitor calpastatin, which is an intrinsically
unstructured protein capable of reversibly binding and inhibiting four molecules of calpain, but
only in the presence of calcium. To date, the mechanism of inhibition by calpastatin and the basis
for its absolute specificity have remained speculative. It was not clear how this unstructured
protein inhibits calpains without being cleaved itself, nor was it known how calcium induced
changes that facilitated the binding of calpastatin to calpain. Here we report the 2.4-A-resolution
crystal structure of the calcium-bound calpain 2 heterodimer bound by one of the four inhibitory
domains of calpastatin. Calpastatin is seen to inhibit calpain by occupying both sides of the
active site cleft. Although the inhibitor passes through the active site cleft it escapes cleavage
in a novel manner by looping out and around the active site cysteine. The inhibitory domain of
calpastatin recognizes multiple lower affinity sites present only in the calcium-bound form of the
enzyme, resulting in an interaction that is tight, specific and calcium dependent. This crystal
structure, and that of a related complex, also reveal the conformational changes that calpain
undergoes on binding calcium, which include opening of the active site cleft and movement of the

Every time I scroll through my list of ‘Friends’ on Facebook, I inevitably come to
the same (somewhat depressing) conclusion: I have absolutely no idea who many of these people
are. This is mostly my fault - my standards have always been pretty low when it comes to
accepting inbound requests, and I never kept up with assigning them to appropriate Friends Lists.
But there’s also the fact that Facebook doesn’t do much to automatically
differentiate between friends and acquaintances (while it does filter your News Feed based on who
it thinks you’re interested in, there isn’t a way to automatically generate a list of
“good friends” versus everyone else).
Meezoog, an Israeli
startup backed by veteran VC Yossi Vardi, is looking to help differentiate between acquaintances
and close friends. Today the company has launched its own social network at Meezoog.com that
attempts to determine how strong the connections are between users by analyzing their
relationships on other sites across the web (it also asks you to manually input your relationship
with each friend, but this isn’t required). The site presents a number of
“paths” connecting you with each friend in a manner similar to Six Degrees of Kevin
Bacon, that helps users determine the degree of “social trust” they have with
each user. At this point it’s too early to tell how well the system works, but it’s a
step in the right direction.
Besides its social network, Meezoog also plans to offer Facebook and OpenSocial applications that
allow users to estimate the nature of their connections with their friends, even if they
aren’t on Meezoog’s network. This cross-network analysis is probably the
company’s best shot at success - while Meezoog may be able to establish its standalone
social network in some regional areas, it will have a very hard time directly competing with
established networks like Facebook and MySpace.
Other companies have tried to do similar social connection strength mapping, including Linking Universe.

Neuroscibr/br/Parkinson's disease (PD) is a progressive neurodegenerative disease characterized by
the degeneration of the dopaminergic neurons in the substantia nigra pars compacta (SNpc).
Activation of the mixed lineage kinase and c-Jun N-terminal kinase (JNK) has been reported in
models of PD. Our focus was to discern whether distinct pathways were activated in cell-specific
manner within the SNpc. We now demonstrate the selective phosphorylation of p38 MAP kinase within
the dopaminergic neurons, whereas JNK activation occurs predominantly in the microglia. p38
activation results in downstream phosphorylation of p53 and increased p53 mediated transcription of
Bax and Puma in the ventral midbrain. Treatment with p38 inhibitor, SB239063 protected primary
dopaminergic neurons derived from human progenitor cells from MPP(+) mediated cell death and
prevented the downstream phosphorylation of p53 and its translocation to the nucleus in vivo, in
the ventral midbrain. The increased staining of phosphorylated p38 in the surviving neurons of SNpc
in human brain sections from patients with PD and in MPTP treated mice but not in the ventral
tegmental area provides further evidence suggesting a role for p38 in the degeneration of
dopaminergic neurons of SNpc. We thus demonstrate the cell specific activation of MAP kinase
pathways within the SNpc after MPTP treatment emphasizing the role of multiple signaling cascades
in the pathogenesis and progression of the disease. Selective inhibitors of p38 may therefore, help

every song with the then inescapable Auto-Tune effect and, less problematically, lean on the
common element — the Roland TR-808 drum machine — of classics like “Make It
Last Forever,” “Posse on Broadway,” “808,” and “Bossy”?
It would have been a wreck, a case of an artist working through paralyzing heartache while loose
in a toy store. Except West wasn’t joking. Not only did he go through with it, but
Roc-A-Fella released the result in time for the 2008 Christmas shopping season. It was indeed a
wreck, if a kind of fascinating one, which helped make the material — voiced by someone who
could not really sing, whose substantial shortcomings were not made less obvious by a polarizing
studio device — seem a little less difficult on the ears.
In various spots across 808s &
Heartbreak, the constant flutter of West’s processed voice, along with a seldom
interrupted sluggish march of aching sounds, is enlivened by the disarming manner in which
despair and dejection are conveyed. When, in “Welcome to Heartbreak,” he
dispassionately recounts sitting alone on a flight, ahead of a laughing family, he makes first
class sound like Siberia; he’d swap lives with the father in an instant. The majority of
the lyrics, however, are directed at an ex who evidently did some damage; in
“RoboCop” alone, she gets compared to the antagonist in Misery and is called
a “spoiled little L.A. girl.” Earlier in the album, the number she did on him is
called “the coldest story ever told,” yet he admits he still fantasizes about her.
All the blocky drums, dragging strings, droning synths, and joyless pianos lead to a bleak set of
productions — even the synthetic calliope in “Heartless” is unnerved, and the
relative pep of “Paranoid” provides no respite, its bitter lyrics subverting a
boisterous beat. Several tracks have almost as much in common with irrefutably bleak post-punk
albums, such as New Order’s Movement and the Cure’s Pornography, as contemporary rap and R&B. (”Coldest Winter,”
where West longs for his departed mother, samples the most desolate song from the first
Tears for Fears album.) For anyone sifting through a broken relationship and self-letdown,
this could all be therapeutic. Otherwise, no matter its commendable fearlessness, the album is a
listless, bleary trudge along West’s permafrost.

receptor-gamma (PPARgamma) signaling after stroke may reduce brain injury, but this effect will
depend on the levels of receptor and cofactors. Here, we showed that the direct effect of PPARgamma
signaling to protect neurons from ischemic injury requires a novel cofactor LMO4, because this
effect was lost in LMO4-null cortical neurons. PPARgamma agonist also failed to reduce cerebral
infarction after transient focal ischemia in CaMKIIalphaCre/LMO4loxP mice with LMO4 ablated in
neurons of the forebrain. Expressing LMO4 in LMO4-null cortical neurons rescued the
PPARgamma-protective effect. PPARgamma signaling activates the promoter of the antioxidant gene
SOD2 and this process requires LMO4. Addition of a superoxide dismutase mimetic MnTBAP
[manganese(III)tetrakis(4-benzoic acid)porphyrin] bypassed the deficiency in PPARgamma signaling
and was able to directly rescue LMO4-null cortical neurons from ischemic injury. Like LMO4,
PPARgamma and PGC1alpha (PPARgamma coactivator 1alpha) levels in neurons are elevated by hypoxic
stress, and absence of LMO4 impairs their upregulation. Coimmunoprecipitation and mammalian
two-hybrid assays revealed that LMO4 interacts in a ligand-dependent manner with PPARgamma. LMO4
augments PPARgamma-dependent gene activation, in part, by promoting RXRalpha (retinoid X
receptor-alpha) binding to PPARgamma and by increasing PPARgamma binding to its target DNA
sequence. Together, our results identify LMO4 as an essential hypoxia-inducible cofactor required
for PPARgamma signaling in neurons. Thus, upregulation of LMO4 expression after stroke is likely to

the integrated response of the CNS to changes in nutritional state. Neurons within the
paraventricular nucleus (PVN) of the hypothalamus express leptin receptors and receive dense
innervation from leptin receptor-expressing neurons in the arcuate nucleus. To obtain new insights
into the effects of circulating leptin on PVN function, we compared global transcriptional profiles
of laser-captured PVN from ad libitum fed mice versus 48 h fasted mice receiving either sham or
leptin treatment intraperitoneally. Five hundred twenty-seven PVN-expressed genes were altered by
fasting in a manner that was at least partially reversible by leptin. Consistent with previous
reports, thyrotrophin releasing hormone mRNA levels were decreased by fasting but restored to fed
levels with leptin treatment. mRNA levels of oxytocin, vasopressin, and somatostatin were also
reduced by fasting and restored by leptin. Given the known effects of leptin on synaptic
remodeling, it is notable that, among the top 15 genes that were positively regulated by leptin,
five have been implicated in synaptic function and/or plasticity (basigin, apolipoprotein E, Gap43,
GABA(A) receptor-associated protein, and synuclein-gamma). Pathway analysis identified oxidative
phosphorylation, in particular, genes encoding complex 1 proteins that play a role in ubiquinone
biosynthesis, to be the predominant gene set that was significantly regulated in a leptin-dependent
manner. Thus, in addition to its effects on the expression of a broad range of neuropeptides,
leptin may also exert more general influences on synaptic function in, and the bioenergetic state
